Defitelio (defibrotide) is an injectable medication used to treat certain complications after a bone marrow transplant, also known as hematopoietic stem cell transplant (HSCT). Defitelio (defibrotide) is infused into the vein (intravenous, IV) by a healthcare provider, and can be given to both children and adults.
Defitelio (defibrotide) is an injectable medication used to manage certain complications after bone marrow transplants, particularly a type of liver problem called hepatic veno-occlusive disease (VOD). Hepatic veno-occlusive disease (VOD), also known as sinusoidal obstruction syndrome (SOS), is a condition where veins in your liver become blocked, which can lead to blood flow problems in the liver and eventually cause liver damage. It's thought to be caused by higher doses of chemotherapy or radiation usually given before the bone marrow transplant to prepare the body for the transplant.
It's not exactly known how Defitelio (defibrotide) works, but it's believed to break down blood clots and protect your cells from damage caused by chemotherapy or radiation.
Your provider will calculate your dose of Defitelio (defibrotide) based on your body weight. It's typically given as a 2-hour IV infusion every 6 hours for at least 21 days. Your provider can continue treatment until signs and symptoms of VOD get resolved, up to a maximum of 60 days.
The most common side effects of Defitelio (defibrotide) include low blood pressure, diarrhea, vomiting, nausea, and nosebleeds. Although rare, some people have experienced an allergic reaction to Defitelio (defibrotide), which can include rash, hives, trouble breathing, and swelling. Get medical help right away if you experience any signs of an allergic reaction.
Because of the way it works, Defitelio (defibrotide) can raise the risk of bleeding. Talk to your provider before starting Defitelio (defibrotide) if you're taking other medications that also raise the risk of bleeding, such as blood thinners. Let your provider know right away if you experience any signs and symptoms of bleeding, such as easy bruising, blood in your urine or stool, headache, confusion, slurred speech, or vision changes. If you develop bleeding, your provider might need to stop Defitelio (defibrotide) and treat the bleeding before starting it again.
